Antioxidative Activity of Flavonoids in Persimmon Leaves

Abstract
The ethylaeetate fraction obtained from the methanol extract of leaves of the persimmon Diospyros kaki Thunb., with strong antioxidative activity, was fractionated by Amberlite XAD-2 column chromatography, and the antioxidative components were separated by Sephadex LH-20 column chromatography and HPLC techniques. As a result, the isolated four flavonoid compounds such as isoquercitrin (quercetin 3-¥Ï-¥â-D-glucopyranoside), quercetin 3-¥Ï-¥â-D-glucopyranoside-2"-gallate, kaempferol 3-¥Ï-¥â-D-glucopyranoside-2"-gallate and astragalin (kaempferol 3-¥Ï-¥â-D-glucopyranoside) were identified by ©öHand ^(13)C-NMR spectroscopy. The antioxidative activity of the four flavonoids was much higher than that of ¥á-tocopherol. In particular, quercetin 3-¥Ï-¥â-D-glucopyranoside-2"-gallate showed the most potent inhibitory activity against the lipid peroxidation of a rabbit erythrocyte membrane ghost, and the, soybean lipoxygenase.
